Chapter 6 - THE CLINICAL INVESTIGATION

The next morning, I didn't go home.
While Brandon stayed at the hospital to watch over Luke, who was finally sitting up and eating a bowl of oatmeal, I drove straight to the university biology labs where I had worked for the past eight years.
My office was a small, quiet space filled with textbooks, stacks of peer-reviewed journals, and a high-powered digital microscope that connected to the university’s main genetic database. I sat at my desk, my fingers flying over the keyboard as I logged into the national medical archives.
There had to be an explanation. In the field of genetics, there are always anomalies—rare, beautiful mistakes of nature that defy the standard high school textbook rules of inheritance.
I spent four hours analyzing Luke’s blood chemistry and comparing it to rare genetic papers from clinics in Europe and Japan.
And then, I found it.
A rare genetic phenomenon known as Cis-AB.
Ordinarily, the A and B genes are located on different chromosomes, meaning an AB parent can only pass down either an A or a B to their offspring, making an O child impossible. But in extremely rare cases—less than 0.001% of the population—the A and B genes are mutated onto the same chromosome. A person with Cis-AB has the blood type AB on paper, but they can pass down an O gene to their child, resulting in a perfectly healthy, biologically related O-negative child.
My heart leaped into my throat. The screen was glowing with the genetic maps of families in Korea and Germany who had faced the exact same crisis—fathers accused of being cuckolded, families destroyed by standard blood tests, only to be saved by a deep-level genetic sequencing.
I grabbed my keys, printed out the research papers, and ran down the hallway to the clinical pathology lab.
“Elena,” I gasped as I burst through the doors of the hospital’s diagnostic wing, finding Dr. Vance sitting at her desk with a cup of coffee. “I need you to run a deep-level PCR sequencing on Brandon’s blood. Don't just do a standard typing. Look for the Cis-AB mutation on the ABO gene.”
Dr. Vance looked up, her eyes widening as she saw the stack of papers in my hands. She took them, scanning the abstracts with the rapid, professional speed of a researcher.
“Cis-AB,” she muttered, her finger tracing a diagram of a mutated chromosome. “It’s extremely rare, Sarah. Most commercial labs don't even have the primer sets to detect it.”
“We have the primers in my lab,” I said, my voice shaking with excitement. “I can bring them over right now. If Brandon has the Cis-AB mutation, it proves he is Luke’s father. It proves the standard blood test was a false negative.”
“Let's do it,” Dr. Vance said, standing up and grabbing a sterile lavender-top tube. “If we can prove this, we can void any claim Sterling is making to the estate court.”
We spent the next six hours in the quiet, sterile warmth of the PCR lab. The machine hummed, heating and cooling the DNA samples we had taken from Brandon’s arm, copying the target sequences millions of times until they were large enough to be read by the digital sequencer.
At 7:00 PM, the monitor flashed with the results.
I leaned close to the screen, my breath catching in my throat.
The green peak on the chromatogram was clear, sharp, and undeniable. Brandon did not have the Cis-AB mutation. His chromosomes were perfectly standard. His A and B genes were on separate alleles, just like 99% of the population.
The genetic path was closed.
Brandon was not, biologically speaking, the father of Luke.
I sat back in the metal lab chair, the cold steel pressing through my lab coat. The room was spinning. The sequencer’s fan hummed a low, mocking note in the quiet room.
If Brandon wasn't Luke’s father... and if I had never been with another man... then how was this possible?
I stared at the screen, my mind desperate, clawing at the edges of reality. And then, a small, dark thought began to take root in the corner of my brain. A thought so wild, so terrifying, that it made my skin turn to ice.
